Development of Molecular Probe for Iron Reducing and Thiosulfate Reducing Bacteria

Microbiologically Influenced Corrosion (MIC) activities are mostly monitored by using canonical culture-based methods. However, these methods detect only culturable bacteria or require months of incubation to get results. To overcome this, molecular detection methods were developed for many of the MIC-causing microbes but were not available for Iron Reducing Bacteria (IRB) and Thiosulfate Reducing Bacteria (TRB). IRB and TRB are among the dominant group of microbes involved in corrosion and souring oil and gas infrastructures. In the present study, molecular probes were developed for IRB and TRB by targeting functional genes implicated in iron and thiosulfate reduction, respectively. Candidate gene sequences were retrieved from genome repository databases of pure isolates or metagenomic sequences. The sequences were aligned using bioinformatic software and primer-based probes were developed from conserved regions. Specificity and efficiency of the primers were validated by amplifying the sequences of target and non-target microbes using Quantitative Polymerase Chain Reaction (qPCR) methods. The probes successfully detected candidate microbes in biocorrosion samples collected from various gas processing facilities. The identified primer-based probes will help oil and gas industries to monitor MIC activities and develop timely management strategies to prevent undesirable microbial activities while improving system integrity and safety concerns.

Effects of Renewable Natural Gas and Hydrogen on Microbially Influenced Corrosion and Souring in Underground Gas Storage

A bench scale laboratory experiment was conducted to assess the microbial corrosion and souring effects renewable natural gas (RNG) and natural gas amended with hydrogen have on underground gas storage systems. Steel coupon-containing vials inoculated with produced fluid from an underground storage well were subject to different headspace conditions to simulate underground gas storage. The vials were filled with geologic natural gas, geologic natural gas amended with hydrogen, RNG, and RNG amended with hydrogen. Each condition was subject to molecular (qPCR, and 16S rRNA analysis), liquid chemistry (sulfate and volatile fatty acid), corrosion, and headspace chemistry (major components and trace sulfur) analysis in duplicate at three time points. The results showed that levels of sulfide production, microbial community composition, and coupon mass loss did not differ greatly between the natural gas and RNG conditions. However, the presence of hydrogen did increase the levels of sulfide in both geologic and RNG conditions. There was also evidence of significant biotransformation of hydrogen into an organic acid (formate). This experiment has shown that microbes native to underground storage wells could have an impact on gas quality and corrosion (via hydrogen sulfide production) if hydrogen is introduced.

The adoption of renewable feedstocks by refiners is driven by a variety of factors, including regulatory compliance, sustainability, and diversification of their product portfolio. However, the introduction of these feedstocks poses significant risks due to their chemical differences compared to traditional crude feedstocks. To address these risks, many refiners have undertaken costly conversion projects to protect their plants from damage caused by these new feedstocks. In addition, refiners are increasingly turning to online integrity sensors to quickly detect corrosion and mitigate unwanted risk, helping to ensure that their critical assets remain healthy and reliable. With these strategies in place, refiners can effectively manage the challenges of incorporating “bio” feedstocks into their refining processes while ensuring the safety and longevity of their equipment. This paper will feature a case study from a major European refiner who have converted an old hydroprocessing unit into producing sustainable fuels from 100% bio feedstock.

The incidence and proliferation of microbial population in oil and gas production facilities can have undesirable consequences on upstream, midstream and downstream production systems. Microbes thrive in the anaerobic conditions encountered in these systems and are supported by nutrients and metabolites found in produced water. Microbiologically influenced corrosion (MIC) is a key oilfield problem associated with microbial activity, and can be described as the accelerated corrosion of surfaces (usually concrete or iron/steel) by the biological action of naturally present or externally introduced microorganisms. Seawater biofouling is a major threat in heat exchanger operations. It decreases the heat transfer efficiency and service life of heat exchangers^1,2. The formation of deposits caused by biofouling on the heat exchanger metal surfaces increases surface roughness and decreases cross-sectional flow area, which leads to higher friction loss in fluid flow^3,4. Mitigation methods, including surface scrubbing, fluidizing bed heat exchangers, cleaning-in-place and dosing anti-fouling chemicals, are the main ways to tackle biofouling⁵. Conventional approaches to treat biofouled components by periodic electrochlorination or acid flushes are costly and environmentally hazardous. Huge costs are associated with heat exchanger biofouling losses, but there is still a lack of research to develop heat-conducting antifouling coatings to heat exchangers³.
